DNA histogram of a mixture of nuclei from leaf tissue of a diploid and a triploid sugar-beet (Partec CyStain UV, UV excitation). The peak positions on the X-axis correspond to the ploidy level, the first peak represents the diploid, the second peak represents the triploid nuclei. The peaks 3 and 4 contain the DNA values of the endomitotic nuclei (4 n, 6 n). The results of the automatic peak evaluations appear immediately after each measurement (peak position, cell number per peak, %, peak width: CV).

Peak positions in the DNA histograms of 50 sugar beet seedlings (100% corresponds to 27 chromosomes: triploid).
8 seedlings of this sample (16%) show aneuploid DNA values (26 resp. 28 chromosomes).

Procedure for fresh or frozen plant material

Ploidy determinations with the Partec PA Ploidy Analyser are perФормаed by the precise measurement of the total DNA content of individual nuclei, which can be derived from almost any plant tissue.
The Ploidy Analyser measures the DNA content of many thousands of individual nuclei within seconds or minutes.
The result is displayed as a DNA histogram in real time on a large LCD screen together with the ploidy level. The ploidy analysis is extremely time and cost efficient.

	approx 0.5 cm square leaf or other plant material
	chop with a sharp razor blade in 0.5 мл of Partec CyStain UV solution A in a plastic petri dish
	filter the sample through a 30 мкл partec CellTrics® directly into the sample tube
	wait 1 to 15 min at room temperature (best time must be determined for each plant variety)
	add at least 2 мл Partec CyStain UV solution B
	connect sample tube to the Ploidy Analyser
	wait approx. 20 sec for optimal DNA histogram

1. PA-I использует лампу HBO-100 "длинная жизнь", встроенный персональный компьютер с программным обеспечением для измерения, анализа результатов и отчетами. Прибор при необходимости оснащается взаимозаменяемым набором фильтров для UV возбуждения и синей эмиссии, другими необязательными фильтрами.

2. Если Вам необходим лазер легкого возбуждения одновременно со свет UV- возбуждения, Partec PA-II позволяет одновременно с лампой HBO-100 "длинная жизнь" применять аргоновую лампу для плучения лазерного света 488 нм. В этой конфигурации можно работать почти со всеми флуоресцентными метками: DAPI, Hoechst, PI, EB, MMC, FITC, FDA и т.д.
